摘要:目的:研究黑莓花色苷提取物对过氧化氢(H:0:)诱导损伤的血管内皮细胞的保护作用及机理。方法:采用倒置相差显微镜观察凋亡细胞的形态学特征,MTI"法观察细胞存活率,试剂盒法测定抗氧化酶活性,流式细胞计法测定细胞凋亡。结果:用不同剂量的黑莓花色苷预处理,细胞形态与损伤组相比较为完整。经50~200肛g/mL黑莓花色苷预处理,内皮细胞存活率由106.5%升至157.0%,显著降低H:O:对血管内皮细胞增殖的抑制作用(P<0.01);黑莓花色苷预处理可显著提高血管内皮细胞过氧化物酶(POD)和超氧化物歧化酶(SOD)活性,200pg/mL黑莓花色苷预处理使POD活性提高50%;而血管内皮细胞的早期凋亡率从12.96%降低到5.21%,晚期凋亡从18.16%降低到2.01%,与H:O:处理比,极显著降低(P<0.01)。结论:黑莓花色苷通过提高细胞存活率,提高细胞抗氧化酶活性,抑制细胞凋亡,对H:0:诱导的血管内皮细胞损伤起到保护作用。
